This vector changes the balance of tRNAs in host cells, thus improving expression of sequences utilizing codons rare used in Escherichia coli cells.
This vector is a small, medium copy number, E.coli plasmid, 4768 bp in length.
The plasmid contains:
1) the p15A replicon (ori) responsible for plasmid replication (source - plasmid pACYC184);
2) the Cm(R) gene, coding for chloramphenicol acetyl transferase, which confers resistance to chloramphenicol (source - plasmid pACYC184);
3) genes for E.coli tRNAs recognizing codons: ATA (Ile-tRNA), GAG (Arg-tRNA), CTA (tRNA2), ATG (tRNA1 and tRNA5), CAA (tRNA3 and tRNA4) and CAG (tRNA6 and tRNA7).
The plasmid is fully compatible with those carrying the pMB1 and ColE1 replicons.
