This vector changes the balance of tRNAs in host cells, thus improving expression of sequences utilizing codons rare used in Escherichia coli cells. Also, it encodes lysozyme of bacteriophage T7 reducing 'leakage" of bacteriophage T7 promoter thus stabilizing persistence of expression plasmids encoding toxic proteins under control of T7 promoter in host cells.

This vector is a medium copy number, E.coli plasmid, 6146 bp in length.

The plasmid contains:
1) the p15A replicon (ori) responsible for plasmid replication (source - plasmid pACYC184);
2) the Spc(R) sequence confers resistance to spectinomycin;
3) genes for E.coli tRNAs recognizing codons: ATA (Ile-tRNA), GAG (Arg-tRNA), CTA (tRNA2), and ATG (tRNA1);
4) the LYSOZYME sequence encode corresponding enzyme of bacteriophage T7.

The plasmid is fully compatible with those carrying the pMB1 and ColE1 replicons.
